Computer Aided Drug Design:
Your responsibility and focus will be on new and challenging innovations in the areas of computational enzyme structure and the understanding of enzyme binding, catalysis, and selectivity in the context of simulations. Possible relevant research experience would include computational enzyme design, applying computational methods (e.g. docking, homology modeling, conformational analysis, molecular dynamics) to the prediction of enzyme structures, binding, catalysis, and selectivity. Knowledge about QM/MM is an essential. Expertise in scientific programming and scripting is a plus. Metabolomics:
Metabonomics study is to profile the complete metabolite levels of cells, tissues and/or a whole organism and their changes as response to a disease (or disease status) or an drug (dosing levels), and eventually to identify disease or drug related differences in one or more of the numerous endogenous metabolites. Combined metabonomic, proteomic and genomic studies (-omics) can lead to accelerated development of mechanistic hypotheses for a disease, and improved understanding of clinical responses to a drug (e.g., efficacy and toxicity) from the level of molecular pathways and regulatory networks. The knowledge acquired from the omics has been integrated into systems biology research; and the omics approaches are being adopted in drug discovery process.

The successful candidate will be involved in a research program aimed at (1) building up an analytical platform for metabolic profiling, and (2) utilizing integrated omics (i.e., genomics, proteomics and metabolomics) approaches to characterize cancer cell lines and cancer stem cells (CSC) to advance our understanding of the relevance of cancer cell metabolism changes associated with cellular proliferation and tumor progression, thus to gain insights into the disease mechanism, and potentially to identify metabolite markers. The researcher will have opportunities of collaborating with internal and external biological and chemical scientists in many aspects.

Tumor Models and Cancer Stem Cells: In developing drugs combating human cancers, tumor models are essential for studying mechanisms of cancer formation, as well as for testing drug candidates. Growing body of evidence suggests the presence of cancer stem cells (CSCs) in tumors. CSCs can be resistant to chemotherapy agents, and only represent a small fraction (2-5%) of cells in a tumor. They posses capacity of self-renewal and are able to differentiate into mature cells. We are performing studies to identify and characterize CSCs. We are also developing novel tumor models for cancer studies and efficacy testing of drug candidates. These studies will facilitate our understanding of growth controls in malignant tumors. In addition, it would serve as model system to understand the basic biology of tumor formation, and, to facilitate discovery and development of therapeutic candidates against cancer (or contact Dr. Z. Stem Cell Biology: Diseases caused liver injury often leads to liver dysfunction and organ failure. Stem cell research offers great promises of applying cell-based therapy and liver regeneration to replace damaged tissue as a means to treat liver diseases. Using integrative approaches including genomics/systems biology, mouse and zebra fish genetics, high content image analysis, chemogenetics, and cutting-edge high through put screening technologies, we are setting up a research program to explore the molecular mechanism underlying stem cell hepatogenic differentiation and liver regeneration. In particular, we are interested in how BMP regulates stem cell pluripotency and hepatogenic lineage progression as well as developing a MTS phenotypic screen to identify molecules that promotes or inhibits stem cell originated hepatocyte differentiation. Virology: Hepatitis B virus is a major health problem in China. We are interested in characterizing the unique process of HBV virus assembly and release from host cells. We will investigate the biological functions of viral envelop proteins and their interaction with cellular factors that regulate the viral assembly and egress pathway. Through the study, we will identify novel antiviral targets and elucidate the mechanisms of HBV life cycle. Virology/epigenetics: Successful treatment of HBV infection hinges on the inactivation or removal of viral replication template which is an episomal, covalently closed and circularized (ccc) double strand DNA (cccDNA) found in cell nucleus. cccDNA is stably maintained as nucleosome attached with H3 and H4 histones. It is shown that acetylation of H3 and H4 histone is related to the activation of viral transcription and replication. We hypothesize that treatment related innate immune activity is able to induce deacetylation of histone proteins in cccDNA nucelosomes and subsequently shuts down viral production. The aim of this study is to investigate the interplays among host innate immune response, deacetylation of cccDNA nucleosome and HBV replication and maintenance. Epigenetics: Epigenetics describe heritable changes in genome function that are not coded in the primary DNA sequence. Many diseases are driven by both genetic and epigenetic abnormalities. Epigenetic alternations in diseases can be reversed, thus represents a new frontier of medicine and will provide innovative therapeutic targets for drug discovery.
