- KYSE150 cells were divided into four groups with three replicates for each group, namely the control group, bufalin treatment group, radiotherapy group, bufalin combined with radiotherapy group. The control group was KYSE150 cells with no treatment. The bufalin treatment group was KYSE150 cells received 80 nM bufalin treatment for 24 h. The radiotherapy group was KYSE150 cells received 4 Gy X-ray irradiation. In the bufalin combined with radiotherapy group, KYSE150 cells were first treated with 80 nM bufalin pretreatment, and then treated with 4 Gy radiotherapy 24h later.
- Total RNA was extracted from KYSE150 cells using Cell Total RNA Extraction Kit (TIANGEN; Beijing, China). RNA integrity was verified by Agilent 4200 Bioanalyzer (CA, USA) and RNA quantification was performed by ND-2000 (Nanodroplet technology). The MGIEasy-rRNA Removal kit (BGI; Shenzhen, China) was used to remove rRNA, and the library was prepared according to the MGIEasy-RNA Library Preparation kit (96RXN). The qualified cDNA library was sequenced by BGI high-throughput sequencing platform, and the original sequencing data were finally obtained.
