ProtocolscRNA-seq libraries were constructed using a DNBelab C Series Single-Cell Library Prep Set (MGI, 940-001924-00). The process includes single cell encapsulation, mRNA capture, reverse transcription, and cDNA amplification in droplet emulsion. Libraries for each sample were indexed and sequenced using a MGISEQ-2000 sequencer (MGI). Raw reads embedded in fastq files were preprocessed using the pipeline of DNBelab C Series HT singlecell analysis software (https://github.com/MGI-tech-bioinformatics/DNBelab_C_Series_HT_scRNA-analysis-software, V1.2). The mouse reference genome GRCm38 and gene annotation (Release M23) downloaded from GENCODE database were used to align the reads and quantify the gene expression. Count matrixes were generated after filtering out low-quality reads with default parameters.
