- Gene expression profile of short-term or long-term prostate cancer cells treated with progesterone

- 1. Abiraterone acetate (AA) is the second-line therapy for castration resistant prostate cancer patients. Progesterone is the most significant hormones increase after AA treatment. Here we treated prostate cancer cell line with progesterone to study the molecular pathways that promote disease progression. VCaP cells were starved for 2 days and stimulated with ethanol, 10nM progesterone, 100nM progesterone, 1nM DHT, 1 μM enzalutamide, 1 μM enzalutamide plus 10nM progesterone, 1 μM enzalutamide plus 100nM progesterone, 1 μM enzalutamide plus 1nM DHT. Gene expression profiles were examined to detail the global program of gene expression treated with progesterone in prostate cancer cells. Biology triplication were used for each treatment. 2. The correlation between progesterone and worse clinical outcome indicated an oncogenic effect of progesterone at a physiological level. Long term treatment of progesterone may result in irreversible alteration in transcriptome in cancer cells. Here we treated LAPC4 with 5 nM progesterone or ethanol for 6 months to generate the Prog_cell and Ctrl_cell. Prog_cell and Ctrl_cell were harvested for RNA extraction and RNA-seq to study the long-term effect of progesterone. Biology triplication were used for each treatment. 3. GATA2 is a potential transcriptional factor involved in estrogen response pathway in prostate cancer. Here we exam the transcriptional changes in Prog_cell by two siRNA of GATA2 versus random siRNA control. Biology triplication were used for each treatment.
