    Description

    Alternative splicing regulation depends on the coupling between transcription and splicing. However, the interaction between alternative splicing and transcription initiation and termination has remained largely unknown due to limited technologies enabling simultaneous measurement. Here, we developed the longPASS approach to accurately identify and quantify transcription start sites (TSS) and polyadenylation sites (PAS) from long-read RNA sequencing data. Using it, we further constructed full-length transcript isoforms to systematically assess the co-regulatory relationship between alternative splicing and TSS/PAS usage. Our finding revealed thousands of TSS/PAS-associated splicing events (TASE) in human cell, representing a pervasive gene regulation mechanism that is consistent across different genetic perturbations and diverse human tissues.

Umbilical cord blood (UCB) has emerged to exhibit effective anti-aging roles. Here, we conducted a perspective, phase 2, single-arm trial of UCB infusion as an adjuvant consolidation therapy in elderly AML patients (ChiCTR-OPC-15006492).Single-cell RNA sequencing (scRNA-seq) identified enhanced anti-tumor and anti-aging properties of UCB, manifested through activation of immune responses and telomere maintenance.

  • PROJ Proteomic and Phosphoproteomic Profiles of Chinese Localized Prostate Cancer

    Previously, we characterized the whole-genomics, epigenomics, and transcriptomic profiles of prostate cancer in China. Here, we performed comprehensive proteomic and phosphoproteomic profiling of 82 paired tumor tissues and matched adjacent normal tissues from 41 Chinese patients with localized prostate cancer. We identified three proteomic subtypes with significant difference in molecular features and clinical prognosis , and found that the proteomic subtypes exhibited similar heterogeneity as the phosphoproteome. Through a biomarker null distribution analysis, we showed that the combination of proteins and phosphosites as biomarkers in prostate cancer has the best ability to predict biochemical recurrence. Then we identified a 13-protein panel that can effectively predict the biochemical recurrence, which was further validated in three independent published datasets. Finally, through integrated multi-omics analysis, we revealed the mechanistic differences underlying different proteomic subtypes, and discovered that the expression and activity of SRSF1 are essential for the malignant characteristics of prostate cancer cells. Our multi-omics data provides valuable resources for understanding the molecular mechanisms of prostate cancer in Chinese cohort, which can be used in the future to develop personalized treatment strategies, and predict prognosis for prostate cancer patients.
