Sediment metagenomic binning |
Metagenomic |
One- to 10-cm-depth sediment samples were collected from the Challenger Deep of Mariana Trench (11.4037°N, 142.3630°E, 10 853 m) during the MV Zhangjian cruise from December 2016 to January 2017. One sediment column was collected using the Hadal Lander equipped with a box corer. After recovering on board, the sediment samples were immediately subsampled using sterile plastic corers and stored at -80°C on board. Sediment cores were thawed and evenly split in to nine fractions at interval per centimeter depth. Total genomic DNA was extracted from 10-20 gram of sediments from each depth fraction, using the FastDNA® SPIN Kit for Soil (MP Biomedicals, USA). Genomic DNA of each sample were fragmented for metagenomic library preparation. Metagenomic library and sequencing were conducted in BGI group (Shenzhen, China) by BGIseq 500 platform, which generated 238.5 Gb of raw reads for the nine metagenomes (23.1 – 36.8 Gb for individual metagenome, with an average value 26.5 Gb). Raw data were deposited in the National Center for Biotechnology Information (NCBI) database and can be accessed under the project ID PRJNA692099. |
