- Cell landscape of IMQ-treated skin in the keratinocyte-specific H2Ab1-deficient mice
- Description
- Although keratinocytes have been believed to exert critical functions in the pathogenesis of psoriasis, the relationship between keratinocytes and other cell types during the development of psoriasis remains elusive. How the MHC-II molecule in the keratinocyte influences the skin environment during the pathogenesis of psoriasis remains unclear. We constructed the epidermis-specific H2-Ab1 knockout mice and H2Ab1 deficiency in the keratinocyte significantly attenuated IMQ-induced inflammatory response. We also performed single-cell RNA-sequencing on the skin tissues of H2Ab1fl/fl mice and K14.H2Ab1fl/fl mice, which revealed the keratinocyte, NK, and DC clusters were significantly reduced in the skin with keratinocyte H2Ab1 deficiency. Therefore, targeting MHC-II could be a therapeutic target for psoriasis.
