Experiment ID Experiment name Experiment type Experiment protocol Date OEX002183 Functional gene amplicon sequencing Metagenomic To further explore the microbial mechanisms behind thermal adaptation, microbial RNA was extracted from 2ml soil slurry sample immediately using the RNeasy PowerSoil Total RNA Kit according to the manufacturer’s instructions (Qiagen, Germany) at the end of 160-day incubation. The final RNA pellet was suspended in 25 µL RNase/DNase-free water. Genome DNA in resuspension was erased and RNA was reverse transcribed into cDNA using the PrimeScript RT Reagent Kits (Takara, Japan). The concentrations of RNA and cDNA were measured on a Qubit 3.0 fluorometer (Thermo Fisher Scientific, USA). cDNA samples were stored at -80℃ for the next step. Amplicon libraries of functional gene (mcrA) gene were prepared for Illumina sequencing with cDNA using a two-stage PCR protocol. 2020-01-06 - Runs
